Albendazole and Avermectin Tablets
The content of albendazole (C₁₂H₁₅N₃O₂S) and avermectin B₁ (B₁ₐ + B₁b) in this product should each be 90.0%–110.0% of the labeled amount.
Description: White or almost white tablets.
[Identification]
(1) Using the solution from the albendazole assay, perform UV-Vis spectrophotometry (Appendix 0401). A maximum absorption peak is observed at 295 nm, and a minimum absorption peak at 277 nm.
(2) In the chromatogram from the avermectin assay, the retention time of the main peak in the test solution should match that of the reference solution.
Tests: Complies with the general requirements for tablets (Appendix 0101).
Assay
Albendazole: Weigh and finely powder 20 tablets. Accurately weigh a portion equivalent to about 20 mg of albendazole, transfer to a 100 mL volumetric flask, add 10 mL of glacial acetic acid, and shake to dissolve. Dilute to volume with ethanol, mix, and filter. Accurately measure 5 mL of the filtrate into another 100 mL volumetric flask, dilute to volume with ethanol, and mix. Using UV-Vis spectrophotometry (Appendix 0401), measure the absorbance at 295 nm. Calculate the content based on the absorption coefficient (E¹%₁cm) of 444 for C₁₂H₁₅N₃O₂S.
Avermectin: Perform high-performance liquid chromatography (HPLC, Appendix 0512).
Chromatographic Conditions: Use a C18 column; mobile phase: methanol-water (85:15); detection wavelength: 245 nm. The theoretical plates for the avermectin B₁ₐ peak should be ≥1500, and resolution between B₁ₐ and B₁b peaks must meet requirements.
Procedure: Weigh and finely powder 20 tablets. Accurately weigh a portion equivalent to about 20 mg of avermectin, transfer to a 100 mL volumetric flask, add methanol, and shake to dissolve. Dilute to volume with methanol, mix, and filter. Inject 10 μL of the filtrate into the HPLC system. Separately prepare and inject a reference standard solution. Calculate the content using the external standard method based on peak areas.
Action and Use: Antiparasitic agent. Used to treat nematodiasis, trematodiasis, cestodiasis, and acariasis in cattle and sheep.
Dosage and Administration: Oral administration (based on avermectin): 0.3 mg per kg body weight for cattle and sheep.
Precautions:
(1) Contraindicated during lactation.
(2) Avermectin is slightly more toxic than ivermectin; use cautiously in sensitive animals.
(3) Avermectin is light-sensitive and rapidly inactivated by oxidation. Store protected from light.
(4) Highly toxic to fish and shrimp. Avoid contamination of water sources with residues, packaging, or animal waste.
Withdrawal Period: 35 days for cattle and sheep.
